Effect of isolated Tricin from arial part of Allium atrovoilaceum Boiss. on proliferation and apoptosis in PC3 cell line

Background: Prostate cancer is the second leading cause of cancer death in men. Tricin is a flavonoid component, which has biological effects. The aim of this study was to investigate the effects of Tricin on cell growth and proliferation and induction of apoptosis on PC3 cell line.
Materials and Methods: Tricin was isolated from the Allium atroviolaceum using column chromatography and recrystallization methods. The chemical structure of isolate was elucidated by spectroscopic techniques including NMR and MS. The PC3 cell line was cultured in DMEM medium with 10% FBS. The cytotoxic effect of Tricin in different concentrations and 48 hours incubation was evaluated by MTT Assay. After that, the cells were treated with Tricin for 48 hours at a concentration of IC50. Apoptosis was evaluated using Annexin-V kit and flow cytometer device.
Results: The viability of cells was decreased by increasing the concentration of Tricin in dose-dependent manner. 50% of the effect of Tricin on the cell death was related to a concentration of 117.5 ± 4.4 μM and 48 hours after treatment. Apoptosis in PC3 cells was not significant after 48 hours treatment with this concentration of Tricin.
Conclusion: Tricin reduced the growth and proliferation of PC3 cells (P<0.05). The rate of induction of apoptosis by Tricin on PC3 cell line was not significant (P>0.05). The results of this study showed that Tricin leads to cell death in the PC3 cell line, but the mechanism of cell death is not apoptosis and more studies are needed in this regard.
