Comparison of Antibiofilm and Antibacterial Effect of Ganoderma Lucidum and Nisin with Chlorhexidine Mouthwash on Streptococcus Mutans

Background and Objective: Various antimicrobial compounds and solutions are used to control oral bacterial infections, particularly those caused by Streptococcus mutans. Due to the increase in their side effects, there is considerable interest in using natural substances and antimicrobial peptides as alternatives to chemical drugs. Therefore, this study aimed to investigate the anti-biofilm and antibacterial properties of Ganoderma lucidum and nisin compared to chlorhexidine mouthwash on S. mutans.
Methods: In this experimental in vitro study, S. mutans strain PTCC 1683 was used. After reviving the standard strain in Brain Heart Infusion (BHI) broth, extracts of G. lucidum and a nisin solution were prepared. The broth microdilution method was used to determine the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of G. lucidum extract, nisin, and chlorhexidine mouthwash. The antibacterial and anti-biofilm effect of the combination of Ganoderma lucidum extract and nisin was determined using the checkerboard method.
Findings: The MIC of G. lucidum extract and nisin on the standard strain of S. mutans is 2.5 mg/ml and 25 μg/ml, respectively, and the MBC of G. lucidum extract and nisin on the standard strain is 2.5 mg/ml and 50 μg/ml, respectively. Also, this bacterium was sensitive to all dilutions of chlorhexidine mouthwash. As a result of the combined use of G. lucidum extract and nisin, the fractional inhibitory concentration index (FICI) did not show a synergistic effect (FICI=2). By reducing the concentration of G. lucidum extract, nisin, and chlorhexidine mouthwash, the ability to inhibit biofilm formation on the studied strain decreased significantly (p<0.05).
Conclusion: The results of this study demonstrate the antimicrobial and anti-biofilm effects of G. lucidum extract and nisin on S. mutans, although they are less effective than chlorhexidine mouthwash.
