Study of FAS rs1800682 Gene Polymorphism in Thyroid Cancer Susceptibility

Background and Objective: Thyroid cancer is the most common endocrine malignancy. Genetic studies have focused on the role of polymorphisms in genes involved in apoptotic pathways and cancer susceptibility risk. FAS is a cell surface receptor that initiates a cell death signaling cascade upon interaction with its ligand (FASL). This study investigated the association of the FAS-670A/G (rs1800682) gene polymorphism with the risk of thyroid cancer in the northwestern region of Iran.
Methods: In this case-control study, 115 patients with thyroid cancer (confirmation of thyroid cancer tissue by a pathologist) and 125 control subjects (healthy individuals without a history of any cancer in themselves or first-degree relatives) were selected. Five milliliters of peripheral blood samples were obtained. DNA extraction was performed using the proteinase K method and the polymorphic genotypes of the FAS gene (rs1800682) were determined using the PCR-RFLP technique and the MvaI restriction enzyme.
Findings: In this study, the genotype frequencies of AA, AG and GG in patients were 41.74%, 30.43% and 27.82%, respectively, and in the control group were 43.2%, 33.6% and 23.2%, respectively. The allele frequencies of G and A in the patient group were 43.04% and 56.96%, respectively, and in the control group were 40% and 60%, respectively. Based on the results, no significant difference was observed between the patient group and the control group in different inheritance models.
Conclusion: The results of the present study showed that the FAS gene polymorphism - 670A/G (rs1800682) cannot be considered as a predisposing factor for thyroid cancer in the studied population in the northwest region of Iran.
