ارتباط فراوانی پروتئین های P53 و P21 با شاخصهای پاتولوژیک آدنوکارسینوم های کولورکتال

سابقه و هدف: سرطان کولورکتال یکی از مهمترین علل مرگ و میر و ناتوانی در جهان است. فقدان عملکرد پروتئین P53 و (P21(WAF1 یکی از مهمترین مکانیسم های تنظیمی دخیل در روند تشکیل سرطان کولورکتال و مؤثر در پیش آگهی و مقاومت درمانی می باشد. این مطالعه به منظور تعیین فراوانی بیان پروتئین P53 و P21 و ارتباط آن ها با شاخص های پاتولوژیک سرطان کولورکتال انجام شد. <br>مواد و روشها: این مطالعه تحلیلی بر روی 70 نمونه بافت تومور بیماران مبتلا به آدنوکارسینوم کولورکتال شهرستان زاهدان که از سال 1389-1382 تحت کولکتومی قرار گرفته بودند، انجام شد. برش های بافتی برای تعیین وضعیت پروتئین P53 و P21 به روش ایمونوهیستوشیمیایی رنگ شده و مورد ارزیابی قرار گرفت. <br>یافته ها: فراوانی بیان پروتئین P53، سی و هفت مورد (9/52%) و فراوانی بیان پروتئین P21، سی و سه مورد (1/47%) بود. بیان بیش از حد پروتئین P53 با شاخص های پاتولوژیک سرطان رابطه ای نداشت. بین بیان پروتئین P21 و درجه تمایز ارتباط معنی داری وجود داشت (02/0=p) به عبارتی بیان این پروتئین در شرایطی با تمایز بهتر با فراوانی بیشتری همراه بود. <br>نتیجه گیری: نتایج مطالعه نشان داد که جهش ژنP53 نقش مهمی در پیشرفت آدنوکارسینوم کولورکتال دارد و تئوری بیان عمده پروتئین P21 در مسیر غیر وابسته به P53، نقش این پروتئین را در سرطان کولورکتال قوت می بخشد.
BACKGROUND AND OBJECTIVE: Colorectal cancer is the major and cause of morbidity and mortality throughout the world. Loss of activity of P53 and P21 (WAF1) proteins that belong to the cell cycle regulation family of protein, seems to be important regulatory mechanism of carcinogenesis in colorectal cancer and affected prognosis and resistance to chemotherapy. P21 expressed in P53-dependent and independent pathway. This study was designed to assess P53 and P21 frequency and their correlation with pathological parameters in colorectal adenocarcinoma.<br>METHODS: This analytical study was done on 70 paraffin preserved colorectal adenocarcinoma samples of patients in Zahedan, Iran who underwent colectomy from 2003 to 2010. Tissue slice was stained and evaluated by method of immunohistochemistry for P53 and P21 protein and evaluated. <br>FINDINGS: P53 was expressed in 37( 52.9%) patients and P21 was expressed in 33(47.1%). No correlation was found between P53 overexpression and P21 with pathological variables. P21 had a statistically significant association with tumor differentiation (p=0.02). The high frequency of P21 was seen in good differentiated tumors.<br>CONCLUSION: The results of the study show that P53 mutation plays an important role in the development of colorectal cancer. No correlation between P21 expression and P53 statue supports the theory that P21 induction occurs mostly in P53 independent pathway in colorectal cancer.
