ارزیابی میزان فراموشی ناشی از اسکوپولامین بر تعداد آستروسیت های شکنج دندانه ای موش صحرایی نر

سابقه و هدف: ارتباط فعالیت سیستم کولینرژیک و رسپتورهای موسکارینی در حافظه و یادگیری از پیش مشخص شده است. از آنجاییکه آستروسیت ها با سیناپس ها تماس دارند، تغییرات ساختاری و مورفولوژیکی قابل توجه ای در هنگام یادگیری پیدا می کنند. این مطالعه به منظور بررسی اثر میزان فراموشی ناشی از اسکوپولامین بر تعداد آستروسیتهای شکنج دندانه ای رت انجام شد. <br>مواد و روشها: این مطالعه تجربی بر روی 48 سر موش صحرایی نر نژاد ویستار با وزن تقریبی 250–200 گرم که در 6 گروه هشت تایی قرار گرفتند، انجام شد. پس از کانول گذاری دو طرفه در ناحیه CA1 هیپوکامپ، رتها در دستگاه یادگیری اجتنابی مهاری آموزش دیدند. بلافاصله تزریق سالین و یا دوزهای مختلف اسکوپولامین (2µg/Rat و 1 و 5/0) به درون (Cornu Amonis, CA1) انجام شد. تست حافظه 24 ساعت بعد از آموزش به صورت اندازه گیری زمان تاخیر ورود به خانه سیاه گرفته شد. 48 ساعت بعد از تست رفتاری، حیوانات تشریح شدند و برشهای عرضی از هیپوکامپ آنها گرفته و با رنگ آمیزی اختصاصی آستروسیت ها (PTAH) رنگ آمیزی شدند. تراکم آستروسیتها در ناحیه شکنج دندانه ای اندازه گیری و مقایسه شد. <br>یافته ها: تزریق اسکوپولامین توانست فراموشی ایجاد کند. در بررسی بافتی تفاوت معنی داری در تعداد آستروسیتها بین گروه کنترل و گروه شم که فقط استرس جراحی را داشتند، دیده شد (05/0>p). افزایش تعداد آستروسیتهای گروههایی که اسکوپولامین دریافت کرده بودند نسبت به گروه کنترل معنی دار نبود. <br>نتیجه گیری: نتایج مطالعه نشان داد که با فراموشی القاء شده توسط اسکوپولامین، تعداد آستروسیتها در شکنج دندانه ای افزایش می یابد.
BACKGROUND AND OBJECTIVE: The relation between cholinergic system and muscarinic cholinergic receptors (mR) associated with learning and memory has been recognized. Because of astrocytes contact to synapses, they are undergoing remarkable structural and morphological changes in learning. The aim of this study was to evaluate amnesia induced by scopolamine (Antagonist mR) on the number of astrocytes in rats dentate gyrus.<br>METHODS: In this experimental study, 48 male Wistar rats, 200-250 gr were used in 6 groups and each group contains 8 rats. The animals were bilaterally implanted with chronic cannulae in the CA1 regions, trained in a step-through type inhibitory avoidance task, and immediately after training, animals received saline or different doses of scopolamine (0.5, 1 and 2 µg/rat, intra-CA1) and tested 24 h after training to measure step-through latency. Then all rats were sacrificed 48h after behavioural test and cross sections were taken from the dorsal hippocampal formation of the right cerebral hemispheres and stained with PTAH (the specific staining for astrocytes). The area densities of the astrocytes was measured in dentate gyrus and compared in the all groups.<br>FINDINGS: Scopolamine injection can induce amnesia. There was a significant difference in astrocyte number between control and sham group which only had surgical stress (p<0.05). Whereas there was no significant increase in astrocyte number of scopolamine received groups compared to control group.<br>CONCLUSION: The results of this study showed that amnesia induced by scopolamine can little increase astrocytes number in the rats dentate gyrus.
