fa مقایسه اثر باکتری‌های استارتر ماست و لاکتوباسیلوس بولگاریکوس بر فعالیت سلول‌های مونونوکلئار خون محیطی بیماران مبتلا به کولیت اولسراتیو میکروبیولوژی Microbiology تجربی Experimental <div style="text-align: justify;"><span style="font-family:times new roman;"><strong><span style="color:blue;">سابقه</span></strong> <strong><span style="color:blue;">و هدف: </span></strong>کولیت اولسراتیو، بیماری‌‌ التهابی روده است که روده بزرگ و راست‌روده را درگیر می‌کند.از آنجائیکه بیماران مبتلا به کولیت اولسراتیو، سطوح بالایی از سایتوکاین‌های پیش التهابی و ضد التهابی ترشح می‌کنند، هدف از این مطالعه مقایسه اثر باکتری‌های استارتر ماست و لاکتوباسیلوس بولگاریکوس بر فعالیت سلول‌های مونونوکلئار خون محیطی بیماران مبتلا به کولیت اولسراتیو می‌باشد.<br> <strong><span style="color:blue;">مواد و روش&shy;ها: </span></strong>این مطالعه‌ تجربی‌آزمایشگاهی روی ۱۰ بیمار مبتلا به کولیت اولسراتیو شهرهای دزفول، اندیمشک و شوش در دو گروه آزمایش و یک گروه کنترل انجام شد. گروه‌‌های آزمایش، شامل کشت مشترک سلول‌های مونونوکلئار و باکتری مورد نظر در رقت‌های ۰/۱ و ۰/۰۱ و زمان‌های ۴۸ و ۷۲ ساعت و کنترل نیز شامل سلول‌های مونونوکلئار بیمار در زمان‌های ۴۸ و ۷۲ ساعت بودند. متغیر <span dir="LTR">IL۱۰</span> و <span dir="LTR">IL۱&beta;</span> توسط الایزا مورد بررسی و مقایسه قرار گرفتند.<br> <strong><span style="color:blue;">یافته ها:</span></strong> در رقت ۰/۱ و زمان ۴۸ ساعت ترشح <span dir="LTR">IL۱&beta;</span> توسط سلول‌های مونونوکلئار تحریک شده با بولگاریکوس ۲۴۹/۶۱<span dir="LTR">&plusmn;</span>۹۴۰/۴ در مقایسه با استارتر ۱۸۱/۱۱<span dir="LTR">&plusmn;</span>۶۶۹/۱۲ (۰/۰۰۴=<span dir="LTR">p</span>) و در ۷۲ ساعت توسط بولگاریکوس ۲۱۳/۳۴<span dir="LTR">&plusmn;</span>۷۹۶/۳ در مقایسه با استارتر ۱۲۸/۴۱<span dir="LTR">&plusmn;</span>۴۶۴/۲۵ (۰/۰۰۰=<span dir="LTR">p</span>)، در رقت ۰/۰۱ و زمان ۴۸ ساعت نیز توسط بولگاریکوس&nbsp;</span><span new="" style="font-family: " times="">۱۹۸/۵۴</span><span style="font-family:times new roman;"><span dir="LTR">&plusmn;</span>۷۴۷/۵ در مقایسه با استارتر ۱۶۳/۸۲<span dir="LTR">&plusmn;</span>۵۲۹/۲۵ (۰/۰۰۵=<span dir="LTR">p</span>) و در ۷۲ ساعت توسط بولگاریکوس ۱۹۲/۵<span dir="LTR">&plusmn;</span>۶۱۷/۴ در مقایسه با استارتر ۱۳۴/۷۸<span dir="LTR">&plusmn;</span>۴۰۸/۶۲ (۰/۰۰۴=<span dir="LTR">p</span>) دارای افزایش معنی‌داری بود. در میزان ترشح سایتوکاین‌ها در هر دو رقت و هر دو زمان بین هر یک از گروه‌های آزمایش با کنترل نیز افزایش معنی‌داری مشاهده شد.<br> <strong><span style="color:blue;">نتیجه گیری: </span></strong><span style="color:windowtext;">نتایج مطالعه نشان داد که </span>لاکتوباسیلوس بولگاریکوس در مقایسه با استارتر با ترشح <span dir="LTR">IL۱&beta;</span> موجب <span style="color:black;">التهاب می‌گردد. بنابراین استارتر نقش بهتری در کاهش التهاب دارد.</span><span style="color:black;"><span style="font-family:2 mitra;"></span></span></span></div> <div style="text-align: justify;"><span style="font-family:times new roman;"><strong>BACKGROUND AND OBJECTIVE:</strong> Ulcerative colitis is an inflammatory bowel disease and involving colon and rectum. Since patients with ulcerative colitis have high levels of pro-inflammatory and anti-inflammatory cytokines, the aim of this study was to compare the effects of starter bacteria of yogurt and lactobacillus boulagaricus on the activity of peripheral blood mononuclear cells in patients with ulcerative colitis.<br> <strong>METHODS:</strong> This experimental-laboratory study was performed on 10 ulcerative colitis patients in Dezful, Andimeshk and Shoosh in two experimental groups and a control group. Experimental groups included co-culture of PBMC and intended bacteria in dilutions of 0.1 and 0.01 and at 48 and 72 hours, and control group including PBMC of the patient at 48 and 72 hours. Variables IL-10 and IL-1&beta;) were measured by ELISA.<br> <strong>FINDINGS:</strong> There was a significant increase in the secretion of IL1&beta; at dilution of 0.1 and 48 hours by PBMC stimulated with bulgaricus 940.4&plusmn;249.61 in comparison with the starter 669.12&plusmn;181.11 (p=0.004) and in 72 hours by bulgaricus 796.3&plusmn;213.34 in comparison with the starter 464.25&plusmn;128.41 (p=0.000), In dilution of 0.01 and 48 hours by bulgaricus 747.5&plusmn;198.54 in comparison with starter 529.25&plusmn;163.82 (p=0.005) and in 72 hours by bulgaricus 617.4&plusmn;192.5 in comparison with starter 408.62&plusmn;134.78 (P=0.004). Also, there was a significant increase in the secretion of cytokines in both dilution and both times between of the experimental groups and control.<br> <strong>CONCLUSION:</strong> The results of the study showed that Lactobacillus bulgaricus causes inflammation in comparison with the starter by IL1&beta; secretion.
