ارزیابی بیان نوترکیب زنجیره سبک توکسین بوتولینوم تیپ A متصل به یک پپتید نفوذ کننده به سلول

<p dir="RTL" style="text-align: justify"><span style="font-family:times new roman"><span style="color:#0000FF"><strong>سابقه و هدف</strong><strong>:</strong></span> توکسین بوتولینوم تیپ <span dir="LTR">A</span> امروزه به طور گسترده ای به صورت تزریقی در درمان برخی اختلالات انقباضی ماهیچههابه کار می رود.این مطالعه به منظور تولیدپروتئین نوترکیب حاصل از اتصال کوالان زنجیره سبک توکسین بوتولینوم تیپ <span dir="LTR">A</span> (<span dir="LTR">BoNT/A</span>) به پپتید <span dir="LTR">TAT<sub>(47-57)</sub></span>وهمچنین شرایط مناسببرای افزایشبیان و تخلیص این پروتئین انجام شد.</span></p>
<p dir="RTL" style="text-align: justify"><span style="font-family:times new roman"><span style="color:#0000FF"><strong>مواد و روش­ ها:</strong></span>در این مطالعه بنیادی توالی نوکلئوتیدی زنجیره سبک توکسین بوتولینوم تیپ <span dir="LTR">A</span> و پپتید <span dir="LTR">TAT</span>از بانک ژنی به دست آمد وسازه ژنی حاوی اینتوالیهاطراحی و ساخته شد. پس از کلون سازی در وکتور بیانی <span dir="LTR">BL21 (DE3)<em>E. coli</em></span> القای بیان با استفاده از <span dir="LTR">IPTG</span> انجام گرفت.سپس شرایط بهینه دمایی و غلظت مناسب <span dir="LTR">IPTG</span>برای بیشترین میزان بیان بر اساس تفاوت در شدت رنگ میان باندهای پروتئینی ژل در<span dir="LTR">SDS PAGE</span>تعیین شد. پروتئین نوترکیببا کمک ستون کروماتوگرافی حاوی نیکل (<span dir="LTR">Ni-NTA</span>) تخلیصشد.</span></p>
<p dir="RTL" style="text-align: justify"><span style="font-family:times new roman"><span style="color:#0000FF"><strong>یافته ها: </strong></span>پروتئین کایمریک حاصل در شرایط معمول بیان (<span dir="LTR">mM IPTG</span> 1و دمای <span dir="LTR">C</span>˚37) به صورت نامحلول تولید می شود. با بهینه سازی شرایط بیان (<span dir="LTR">mM IPTG</span> 5/0و دمای <span dir="LTR">C</span>˚18)میزان 60% ازپروتئین کایمریک به صورت محلول تولید شد.</span></p>
<p style="text-align: justify"><span style="font-family:times new roman"><span style="color:#0000FF"><strong><span dir="RTL">نتیجه گیری: </span></strong></span><span dir="RTL">بر اساس نتایج این مطالعه با بیان پروتئین نوترکیب به صورت محلول پروتئین پیچش صحیح و در نتیجه فعالیت خود را به میزان زیادی حفظ کرده و نیازی به استفاده از ترکیبات دناتوره کننده برای باز کردن پیچش پروتئین و محلول سازی آن نخواهد بود. </span></span></p>
<p><span style="font-family:times new roman"><span style="color:#0000FF"><strong>BACKGROUND AND OBJECTIVE: </strong></span>Botulinum toxin type A (BoNT/A) is widely used in the treatment of some muscle contraction disorders through injection. This study aimed to produce recombinant protein through covalent bonding of the light chain of BoNT/A to the peptide TAT (47-57), and to create an optimized condition for expression and purification of the protein.</span></p>
<p><span style="font-family:times new roman"><span style="color:#0000FF"><strong>METHODS: </strong></span>In this preliminary study, the nucleotide sequences of the light chain of BoNT/A and TAT peptide were obtained from Gen Bank, and genetic structures containing these sequences were designed and engineered. After cloning into the BL21 (DE3) E. coli vector, expression was induced by IPTG. Thereafter, optimum thermal condition and IPTG concentration for maximum expression were determined based on the difference in the intensity of staining between the bands on SDS-PAGE protein gel. The recombinant protein was purified through nickel column chromatography (Ni-NTA).</span></p>
<p><span style="font-family:times new roman"><span style="color:#0000FF"><strong>FINDINGS:</strong> </span>The produced chimeric protein is insoluble in normal conditions (1 mM IPTG and at 37˚ C). Through optimization of expression conditions (0.5 mM IPTG and at 18˚ C) 60% of the chimeric protein was produced as solution.</span></p>
<p><span style="font-family:times new roman"><span style="color:#0000FF"><strong>CONCLUSION:</strong> </span>Based on our results, through expressing the recombinant protein as a solution, the protein maintained its proper folding and function. Hence, the use of denaturation compounds for solution making and destabilization of folded protein structures is not required. </span></p>
