fa بررسی تاثیر هیپوکسی بر تغییرات بیان 107‏miR-‎‏ در رده های سلولی سرطان معده ‏‏45‏MKN‏ و ‏AGS An Investigation of the Effect of Hypoxia on Expression of 107-miR in ‎Gastric Cancer Cell Lines MKN-45 and AGS‎ ژنتیک، بیولوژی سلولی و مولکولی Genetics, Cell and Molecular Biology تحقیقی Research سابقه و هدف: هیپوکسی در تومورهای جامد، علت اصلی مقاومت به درمان در سرطان است. بنابراین شناخت اندیکاتورهای مناسب جهت آگاهی از وضعیت هیپوکسی در تومور برای انجام روش های صحیح درمانی و آگاهی از پیش اگهی تومور بسیار مفید و ضروری است. هدف از انجام این تحقیق بررسی تغییر بیان miR-107 به عنوان بیومارکر و اندیکاتور هیپوکسی و ارتباط آن با هیپوکسی در سلول های سرطان معده می باشد.<o:p /> مواد و روشها: در این تحقیق، رده سلولی سرطان معده 45MKN و AGS از انستیتو پاستور تهران تهیه و درشرایط اکسیژن نرمال(95% O2 و 5% CO2) و هیپوکسی (95% N2 و 5% CO2) کشت داده شدند و در فواصل زمانی 24 و 48 ساعت بررسی بیان 107miR- و HIF-1α انجام گرفت.<o:p /> یافتهها: با القای هیپوکسی در رده های سلولی بیان HIF-1α در رده 45MKN با افزایش مدت زمان تیمار (24و48 ساعت)، به ترتیب 2/3 و 3/8 برابر افزایش پیدا کرد (0/002 p= و p=0/04) و در رده AGS 2/2 و 3/8 برابر افزایش یافت (0/005 p= و 0/03 p=). همچنین بیان افزایش یافته miR-107 در رده 45MKN در این بازه های زمانی به ترتیب 2/2 و 3/1 برابر افزایش مشاهده شد (0/0001 p= و 0/01 p=) و در رده AGS به ترتیب 2/4 و 4 برابر افزایش داشت (0/0001 p= و 0/002 p=).<o:p /> نتیجه گیری: نتایج مطالعه نشان داد که القای هیپوکسی در رده های سلولی سرطان معده بر میزان تغییرات بیان miR-107 موثر است.<o:p /> BACKGROUND AND OBJECTIVE: Hypoxia in solid tumors is the major cause of cancer treatment resistance. Thus, identifying the appropriate indicators of hypoxia in tumors is of great importance for appropriate tumor prognosis and choosing the best treatment methods. This study aims to investigate the modulation of miR-107 expression, as a biomarker of hypoxia and its association with gastric cancer cells.<o:p /> METHODS: MKN45 and AGS gastric cancer cells were purchased from Tehran Pasteur Institute and then, were cultured under normal oxygen conditions (CO2 5% and O2 95%) and hypoxia (CO2 5% and N2 95%). Finally, miR-107 and HIF-1-α expressions were evaluated every 24 to 48 hours.<o:p /> FINDINGS: The results of the study showed that through hypoxia induction, HIF-1-α expression in MKN-45 cell lines increased by extending treatment duration (24 and 48 hours) 2.3 and 3.8 times, respectively, (p=0.04 and p=0.002), and in AGS cell lines HIF-1-α expression increased 2.2 and 3.8 times (p=0.03 and p=0.005).
