fa بررسی اثر عصاره هیدروالکلی گل میخک (Dianthus deltoides) در پاسخ التهابی با استفاده از تست گزیلن و واکنش به محرک درد به روش آزمون Hot Plate در بین موش های سوری Effects of the Hydro-alcoholic Extract of Clove (Dianthus deltoides) on Inflammation and Pain Response using the Xylene Test and Hot Plate Test in Mice فارماکولوژی Pharmacology مداخله ای Interventional سابقه و هدف: گل میخک حاوی فنلی به نام اوژنول می باشد که دارای خاصیت آنتی اکسیدانی، ضد دردی و ضد التهابی می باشند. لذا این مطالعه به منظور بررسی اثر عصاره هیدروالکلی گل میخک بر التهاب و واکنش به محرک درد در موش سوری انجام شد. مواد و روشها: در این تحقیق تجربی برای بررسی اثر عصاره بر التهاب، از 48 سر موش سوری نر که به 6 گروه شامل گروه شاهد (سالین)، کنترل مثبت (دگزامتازون) و گروه های تجربی (دریافت عصاره به ترتیب با دوزهای 42، 85، 170 و 340 میلی گرم بر کیلوگرم) تقسیم شدند، استفاده گردید. برای بررسی میزان احساس درد، از 160 سر موش سوری نر استفاده شد که به 4 دسته تقسیم و هر دسته به 5 گروه تقسیم شدند که شامل گروه شاهد (سالین)، کنترل مثبت (مرفین)، گروه دریافت کننده عصاره با دوز 200 میلی گرم بر کیلوگرم، گروه دریافت کننده عصاره با دوز 500 میلی گرم بر کیلوگرم و گروه دریافت کننده توام عصاره 200 میلی گرم بر کیلوگرم و نالوکسان (4 میلی گرم بر کیلوگرم) بود. برای بررسی پاسخ به درد از هرکدام از 4 دسته به صورت جداگانه در یکی از زمان های 5، 15، 30 یا 60 دقیقه پس از تزریق، آزمون صفحه داغ گرفته شد. وزن برش های 7 میلی متری تهیه شده از دو گوش پس از دو ساعت از تزریق 03/0 میلی لیتر گزیلن به پشت گوش راست موش ها، با هم مقایسه شدند. یافته ها: در آزمون التهاب در گروه شاهد و گروه دریافت کننده عصاره mg/kg  42 ، دارای بیشترین اختلاف وزن بین برش های تهیه شده از دو گوش حیوان (به ترتیب 1/5±36 و 3/5±33 میکروگرم) بود و کمترین میزان مربوط به دوز   180و 340 mg/kg (به ترتیب 1/2±21 و 1/5±17میکروگرم) بود. در آزمون صفحه داغ، در بین گروه های تیمار شده با عصاره، عصاره mg/kg 500 در 15 دقیقه قبل از آزمون، دارای بیشترین زمان تاخیر و کمترین میزان مربوط به گروه دریافت کننده توام نالوکسان و عصاره mg/kg 200  در 5 دقیقه قبل از آزمون بود (برحسب ثانیه به ترتیب برابر با 1/53±18/91 و 1/04±8/71). نتیجه گیری: نتایج مطالعه نشان داد که عصاره گل میخک دارای خواص ضد التهابی و ضد دردی می باشد. BACKGROUND AND OBJECTIVE: Clove contains large proportion of a phenolic compound called eugenol, which has antioxidant, anti-inflammatory and analgesic properties. This study aimed to evaluate the effects of hydro-alcoholic clove extract on inflammation and pain response in mice.  METHODS: In this experimental study, 48 male mice were classified into six groups of control (saline), positive control (dexamethasone) and treatment (receiving 42, 85, 170 and 340 mg/kg of clove extract) to investigate the effects of clove extract on inflammation. To evaluate pain perception, 160 mice were categorized into 4 groups, divided into 5 subgroups including the control (saline), positive control (morphine), recipients of 200 mg/kg clove extract, recipients of 500 mg/kg of extract, and recipients of 200 mg/kg of combined clove extract and naloxone (4 mg/kg). To assess pain response, the hot plate test was performed on each subgroup at 5, 15, 30 and 60 minutes after the injection of the herbal extract. In addition, two hours after the injection of 0.03 ml xylene into the back of the right ears of the mice, the sections obtained from both ears (7 mm) were compared in terms of weight. FINDINGS: In the inflammation test, the most significant difference in the ear sections was observed between the control group and recipients of 42 mg/kg of extract (36±5.1 and 33±5.3 µg, respectively), while the least significant difference was observed at doses of 180 and 340 mg/kg (21±2.1 and 17 ±1.5 µg, respectively). In the hot plate test on the treatment groups, dose of 500 mg/kg caused the highest pain delay 15 minutes before the test, while the lowest delay was observed in the recipients of combined naloxone and clove extract (200 mg/kg) 5 minutes before the test (18.91±1.53 and 8.71±1.04 seconds, respectively).
