fa اثر عصاره هیدروالکلی گیاه پنج انگشت (vitex agnus custes) بر پیامد سکته مغزی در موش های سوری اوارکتومی شده The Effects of the Ethanolic Extract of Vitex Agnus Castus on Stroke Outcomes in Ovariectomized Mice فیزیولوژی Physiology تحقیقی Research <div style="text-align: right"> <ul> <li style="direction: rtl"> سابقه و هدف: شیوع سکته مغزی در زنان قبل از یائسگی کمتر از مردان است که استروژن به عنوان یک عامل محافظ نورونی قوی در ایسکمی مغزی در سال های پیش از یائسگی شناخته شده است. فیتواستروژن ها خطرات کمتری نسبت به استروژن های شیمیایی دارند. از آنجائیکه گیاه پنج انگشت (vitex agnus custes) دارای خاصیت فیتواستروژنی می باشد در این مطالعه اثر عصاره هیدروالکلی این گیاه بر پیامد سکته مغزی در مدل انسداد دائم شریان مغزی میانی در موش های سوری اوارکتومی شده مورد بررسی قرار گرفت. مواد و روشها: در این مطالعه تجربی، 32 سر موش سوری ماده در محدوده وزنی 25 تا 35 گرم به طور تصادفی به 4 گروه (8 تایی) شم، کنترل (اوارکتومی، یک ماه گاواژ سالین بادوز  ml/kg1 و سپس القاء سکته)، عصاره گیاه (اوارکتومی، گاواژ عصاره گیاه پنج انگشت با دوز  mg/kg80 (در یک میلی لیتر سالین) به مدت یک ماه هر روز) و گروه استروژن (اوارکتومی، گاواژ استرادیول والرات µg/kg40 (در یک میلی لیتر سالین) به مدت یک ماه هر روز) تقسیم شدند. حیوانات اوارکتومی شده و بعد از گذشت یک ماه سکته مغزی به وسیله سوزاندن شریان مغزی میانی القاء شد. حجم انفارکتوس و اختلالات نورولوژیک یک هفته بعد از القاء سکته مغزی اندازه گیری شد. یافته‌ها: حجم سکته در گروه کنترل، استروژن و عصاره گیاه پنج انگشت در یک هفته پس از ایجاد سکته مغزی به ترتیب 33/2±98/11، 01/1±41/3و 10/1±5 درصد بود. هر دو عصاره گیاه پنج انگشت و استروژن، حجم سکته مغزی را نسبت به گروه کنترل کاهش دادند (05/0p<). همچنین عصاره گیاه و استروژن اختلالات نورولوژیک را نسبت به گروه کنترل کاهش دادند (001/0p<). اختلالات حسی حرکتی در گروه های کنترل، استروژن و عصاره گیاه پنج انگشت در یک هفته بعد از سکته مغزی به ترتیب برابر با 7± 50، 2±15 و 09/4±21 بود و یک اختلاف معنی داری را بین گروه کنترل و دیگر گروه ها نشان داد (001/0p<). نتیجه گیری: نتایج مطالعه نشان داد که عصاره گیاه پنج انگشت مشابه با استروژن اثرات محافظت نورونی را به دنبال انسداد دائم شریان مغزی میانی در موش های سوری اوارکتومی شده دارد. </li> </ul> </div> <pre style="line-height: 16pt background: white"> BACKGROUND AND OBJECTIVE: The prevalence of stroke in premenopausal women is lower than men. In fact, estrogen is known as a potent neuroprotective agent in cerebral ischemia before menopause. Phytoestrogens are considered to be less risky than chemical estrogens. In this study, considering the phytoestrogenic properties of Vitex agnus custus, the effects of the ethanolic extract of this plant on stroke outcomes in a model of middle cerebral artery occlusion were investigated in ovariectomized mice.</pre> METHODS: In this experimental study, 32 mice, weighing 25-35 g, were randomly divided into 4 groups (8 mice per group): 1) sham group, 2) control group (ovariectomized, treated with 1 ml/kg saline for one month, followed by stroke induction), 3) Vitex group (ovariectomized, treated with 80 mg/kg of Vitex extracts in 1 mL saline every day for a month), and 4) estrogen group (ovariectomized, treated with 40 μg/kg of estradiol valerate in 1 mL saline every day for a month). After one month, stroke was induced in ovariectomized mice by cauterizing the middle cerebral artery. Infarct volume and neurological disorders were evaluated one week after stroke induction. FINDINGS: A week after stroke induction, infarct volume in the control, estrogen, and Vitex groups was 11.98±2.33, 3.41±1.01, and 5±1.10, respectively. Vitex extracts and estrogen could decrease infarct volume, compared to the control group (p<0.05). Also, estrogen and Vitex extracts reduced neurological deficits, compared to the control group (p<0.001). A week after stroke induction, sensorimotor disorders in the control, estrogen, and Vitex groups were 50±7, 15±2, and 21±4.09, respectively. In fact, a significant difference was observed between the control and other groups (p<0.001). CONCLUSION: The findings of the present study showed that Vitex extracts, similar to estrogen, have neuroprotective effects, following middle cerebral artery occlusion in ovariectomized mice. گیاه پنج انگشت, اوارکتومی, شریان مغزی میانی, سکته مغزی مدل دائم, عصاره گیری.
